Cardiac fibrosis is a common manifestation of various heart diseases, including myocardial infarction, hypertension, diabetes, and ischemia/reperfusion (I/R). The present study aimed to investigate the role of retinoic acid (RA) in the regulation of myocardial fibrosis in the heart. Myocardial fibrosis in C57BL/6 mice was induced by chronic I/R or isoproterenol injection. The effect of RA was studied by intraperitoneal injection of RA (60 mg/kg/day) for 14 days in C57BL/6 mice with myocardial fibrosis. Myocardial fibrosis was examined by Masson staining and α-smooth muscle actin (α-SMA) immunohistochemistry. The expression of α-SMA, tumor necrosis factor-α (TNF-α), transforming growth factor-β1 (TGF-β1), and connective tissue growth factor (CTGF) was detected by western blot and qRT-PCR. Activation of the AMP-activated protein kinase (AMPK) pathway was determined by western blot. RA significantly reduced myocardial fibrosis in mice after chronic I/R or isoproterenol injection. Furthermore, RA markedly inhibited the expression of α-SMA, TNF-α, TGF-β1, and CTGF in the heart of mice with myocardial fibrosis. Moreover, RA upregulated the expression of AMPK and increased its activation in vivo.
